== Function == Trypsinogen is the proenzyme precursor of trypsin. Trypsinogen (the inactive form) is stored in the pancreas so that it may be released when required for protein digestion. The pancreas stores the inactive form trypsinogen because the active trypsin would cause severe damage to the tissue of the pancreas. Trypsinogen is released by the pancreas into the second part of the duodenum, via the pancreatic duct, along with other digestive enzymes.

== History == Providence Therapeutics Holdings Inc. was co-founded in Toronto, Ontario by Calgary, Alberta-based businessman Brad T. Sorenson and San Francisco–based Eric Marcusson Ph.D., who was also the Chief Scientific Officer. PTX-COVID19-B is a messenger RNA (mRNA)-based COVID-19 vaccine. In an interview with CTV news, Sorenson said they were "building some of the important building blocks for the messenger RNA ... that provides instructions to cells ... to build proteins that may treat or prevent disease". As of January 2021, Northern RNA's Calgary lab was proposed as the site where manufacturing of PTX-COVID19-B would take place. A skeletal survey is useful to confirm the diagnosis of achondroplasia. The skull is large, with a narrow foramen magnum, and relatively small skull base. The vertebral bodies are short and flattened with relatively large intervertebral disk height, and there is congenitally narrowed spinal canal. The iliac wings are small and squared, with a narrow sciatic notch and horizontal acetabular roof. The tubular bones are short and thick with metaphyseal cupping and flaring and irregular growth plates. Fibular overgrowth is present. The hand is broad with short metacarpals and phalanges, and a trident configuration. The ribs are short with cupped anterior ends. If the radiographic features are not classic, a search for a different diagnosis should be entertained. Because of the extremely deformed bone structure, people with achondroplasia are often "double jointed". The diagnosis can be made by fetal ultrasound by progressive discordance between the short femur length and biparietal diameter by age. The trident hand configuration can be seen if the fingers are fully extended. Another common characteristic of the syndrome is thoracolumbar gibbus in infancy.

== Synthesis == [18F]FDG was first synthesized via electrophilic fluorination with [18F]F2. Subsequently, a "nucleophilic synthesis" was devised with the same radioisotope. As with all radioactive 18F-labeled radioligands, the fluorine-18 must be made initially as the fluoride anion in a cyclotron. Synthesis of complete [18F]FDG radioactive tracer begins with synthesis of the unattached fluoride radiotracer, since cyclotron bombardment destroys organic molecules of the type usually used for ligands, and in particular, would destroy glucose. Cyclotron production of fluorine-18 may be accomplished by bombardment of neon-20 with deuterons, but usually is done by proton bombardment of 18O-enriched water, causing a (p-n) reaction (sometimes called a "knockout reaction" – a common type of nuclear reaction with high probability where an incoming proton "knocks out" a neutron) in the 18O. This produces "carrier-free" dissolved [18F]fluoride ([18F]F−) ions in the water. The 109.8-minute half-life of fluorine-18 makes rapid and automated chemistry necessary after this point. Anhydrous fluoride salts, which are easier to handle than fluorine gas, can be produced in a cyclotron. To achieve this chemistry, the [18F]F− is separated from the aqueous solvent by trapping it on an ion-exchange column, and eluted with an acetonitrile solution of 2,2,2-cryptand and potassium carbonate. === Heart failure === One of the safety concerns identified before approval was fluid retention. Moreover, the combination of rosiglitazone with insulin resulted in a higher rate of congestive heart failure. In Europe there were contraindications for use in heart failure and combination with insulin. A meta analysis of all trials from 2010 and 2019 confirmed a higher risk of heart failure and a double risk when rosiglitazone was administered as add-on therapy to insulin. Two meta-analyses of real life cohort studies found a higher risk of heart failure compared to pioglitazone. There were 649 excess cases of heart failure every 100,000 patients who received rosiglitazone rather than pioglitazone.

==== Neonatal hypoglycemia ==== Low blood sugar may occur in healthy neonates aged less than 48 hours who have not eaten for a few hours. During the 48-hour neonatal period, the neonate adjusts glucagon and epinephrine levels following birth, which may trigger transient hypoglycemia. In children who are aged greater than 48 hours, serum glucose on average ranges from 70 to 100 mg/dL (3.9–5.5 mmol/L), similar to adults, with hypoglycemia being far less common.
