N-acetyl Semax is one of those subjects where the details matter more than the headlines. This page pulls together the background, the mechanisms, and the practical points readers ask about most.

Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.
Published studies examine a fairly narrow set of endpoints. Rodent experiments commonly measure maze learning, infarct volume after induced ischemia, and tissue levels of neurotrophic factors. Clinical reports from Russian centres describe attention, memory and recovery scores in patients after stroke or transient ischemic attack. Most of those human studies are small and few have been repeated by independent groups. Outcome measures differ between studies, which limits direct comparison.
| Property | Value | Notes |
|---|---|---|
| Primary research models | Rodent studies | Largest share of published data |
| Reported markers | BDNF and NGF expression | Measured mainly in animal tissue |
| Common administration | Intranasal | Matches the registered formulation |
| Blood residence time | Minutes | Rapid enzymatic degradation |
| Evidence quality | Small trials, limited replication | Noted repeatedly in reviews |
Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
=== Metalloprotein–ligand interactions === The primary sequences of thousands of olfactory receptors are known from the genomes of more than a dozen organisms: they are seven-helix transmembrane proteins, but there are very few solved structures. Their sequences exhibit typical class A GPCR motifs, useful for building their structures with molecular modeling. Golebiowski, Ma and Matsunami showed that the mechanism of ligand recognition, although similar to other non-olfactory class A GPCRs, involves residues specific to olfactory receptors, notably in the sixth helix. There is a highly conserved sequence in roughly three quarters of all ORs that is a tripodal metal ion binding site, and Suslick has proposed that the ORs are in fact metalloproteins (mostly likely with zinc, copper and possibly manganese ions) that serve as a Lewis acid site for binding of many odorant molecules. Crabtree, in 1978, had previously suggested that Cu(I) is "the most likely candidate for a metallo-receptor site in olfaction" for strong-smelling volatiles which are also good metal-coordinating ligands, such as thiols. Zhuang, Matsunami and Block, in 2012, confirmed the Crabtree/Suslick proposal for the specific case of a mouse OR, MOR244-3, showing that copper is essential for detection of certain thiols and other sulfur-containing compounds. Thus, by using a chemical that binds to copper in the mouse nose, so that copper was not available to the receptors, the authors showed that the mice could not detect the thiols.
=== Marxist criticisms === Adorno posits social totality as an automatic system. According to Horst Müller's Kritik der kritischen Theorie ("Critique of Critical Theory"), this assumption is consistent with Adorno's idea of society as a self-regulating system, from which one must escape (but from which nobody can escape). For him, it was existent but inhuman. Müller argues against the existence of such a system and claims that critical theory provides no practical solution for societal change. He concludes that Jürgen Habermas, in particular, and the Frankfurt School in general, misconstrue Marx.
Caesium-137 has been used as a tracer in hydrologic studies, analogous to the use of tritium. Small amounts of caesium-134 and caesium-137 were released into the environment during nearly all nuclear weapon tests and some nuclear accidents, most notably the Goiânia accident and the Chernobyl disaster. As of 2005, caesium-137 is the principal source of radiation in the zone of alienation around the Chernobyl nuclear power plant. Its chemical properties as one of the alkali metals make it one of the most problematic of the short-to-medium-lifetime fission products because it easily moves and spreads in nature due to the high water solubility of its salts, and is taken up by the body, which mistakes it for its essential congeners sodium and potassium.
Adding to absinthe's negative reputation in the late 19th and early 20th centuries, unscrupulous makers of the drink omitted the traditional colouring phase of production in favour of adding toxic copper salts to artificially induce a green tint. This practice may be responsible for some of the alleged toxicity historically associated with this beverage. Many modern-day producers resort to other shortcuts, including the use of artificial food coloring to create the green color. Additionally, at least some cheap absinthes produced before the ban were reportedly adulterated with poisonous antimony trichloride, reputed to enhance the louching effect.
== Other species == Humanin is the most well-conserved of the mitochondria-derived peptides, found in such diverse species as humans, naked mole rats, and nematodes. Overexpression of humanin in Caenorhabditis elegans has been shown to extend the lifespan of that nematode by increasing autophagy. The rat, Rattus norvegicus, has a gene, rattin (C0HLU6, "Humanin-like protein"), that encodes a 38 amino acid peptide homologous to humanin. The two genes produce cDNAs that show 88% sequence identity. The peptides are 81% identical, with the carboxyl terminal sequence in rattin being 14 amino acids longer than in humanin. Of the 24 amino acids in the rest of the rat sequence, 20 are identical to the amino acids in the human sequence. The mouse MT-RNR2 humanin ortholog is a pseudogene, so no humanin is produced from the mtDNA. However, the nuclear genome harbors (like in humans) many copies of mitochondrial genomes, and one copy of the humanin homolog, Gm20594 (J3QJY3), is actively expressed.

== Medical uses == Vardenafil's indications and contraindications are the same as with other PDE5 inhibitors; it is closely related in function to sildenafil citrate (Viagra) and tadalafil (Cialis). The difference between the vardenafil molecule and sildenafil citrate is a nitrogen atom's position and the change of sildenafil's piperazine ring methyl group to an ethyl group. Tadalafil is structurally different from both sildenafil and vardenafil. Vardenafil's relatively short effective time is comparable to but somewhat longer than sildenafil's. Despite the structural differences between vardenafil and sildenafil, they both show similar ocular side effects due to their similar selectivity overlap with PDE6, an enzyme found in the eye. Beyond its indications for erectile dysfunction, vardenafil may be effective in the treatment of premature ejaculation, where it may significantly increase the time from penetration to ejaculation.
== Epidemiology == Numerical values regarding prevalence of uterine prolapse differ based on whether the epidemiologic study in question uses a physical exam or a symptom questionnaire to determine the presence of prolapse. Prevalence of pelvic organ prolapse was found to be consistently higher when physical exam was used (for uterine prolapse, this was 14.2% in one study and 3.8% in another) compared to a symptom-based determination in which the prevalence of any type of prolapse, including uterine prolapse, was 2.9% to 8% in the U.S. Using Women's Health Initiative data, the incidence of grades 1 to 3 uterine prolapse was approximately 1.5/100 women-years and progression of uterine prolapse was found to be about 1.9%.

== Mating == Researchers examined whether female beetles preferred males infected by the tapeworm Hymenolepis diminuta as mates, or noninfected males. Virgin females were found to spend more time near — and to copulate more often with — noninfected males in the experiment. Mating behavior was also found to be influenced by the mass of the male beetle. A larger male was preferred and mated with first compared to a smaller male mate. Overall, female beetles showed mating preferences for noninfected males who were larger in size. One explanation provided by researchers for this finding is that larger, noninfected males may allow females to gain genetic or material benefits.
== Overdose == A single oral dose of bicalutamide in humans that results in symptoms of overdose or that is considered to be life-threatening has not been established. Dosages of up to 600 mg/day have been well tolerated in clinical trials, and it is notable that there is a saturation of absorption with bicalutamide such that circulating levels of its active (R)-enantiomer do not further increase above a dosage of 300 mg/day. Overdose is considered unlikely to be life-threatening with bicalutamide or other first-generation NSAAs (i.e., flutamide and nilutamide). A massive overdose of nilutamide (13 grams, or 43 times the normal maximum 300 mg/day clinical dosage) in a 79-year-old man was uneventful, producing no clinical signs, symptoms, or toxicity. There is no specific antidote for bicalutamide or NSAA overdose, and treatment should be based on symptoms, if any are present.

==== PROSITE pattern notation ==== The PROSITE notation uses the IUPAC one-letter codes and conforms to the above description with the exception that a concatenation symbol, '-', is used between pattern elements, but it is often dropped between letters of the pattern alphabet. PROSITE allows the following pattern elements in addition to those described previously:
Phenethylamines, as well as amphetamines (α-methylphenethylamines), are derivatives of β-phenethylamine and are structurally related to the monoamine neurotransmitters dopamine, norepinephrine, and epinephrine. Some phenethylamines and amphetamines, particularly those with methoxy and other substitions on the phenyl ring, are potent serotonin 5-HT2 receptor agonists, including of the serotonin 5-HT2A receptor, and can produce psychedelic effects. In contrast to phenethylamines and amphetamines generally, most psychedelic phenethylamines are not monoamine releasing agents. Examples of psychedelic phenethylamines and amphetamines include mescaline and other scalines like trimethoxyamphetamine (TMA) and escaline, the 2C drugs like 2C-B, 2C-E, and 2C-I, the DOx drugs like DOM, DOB, DOI, certain MDxx drugs like MDA and MDMA (weak psychedelics), FLY drugs like 2C-B-FLY and Bromo-DragonFLY, and the NBOMe (25x-NBx) drugs like 25I-NBOMe, among others.
He was a consistent performer in an inconsistent season for the Bombers, polling 16 Brownlow votes from a total of 43 received by Essendon players, including a three-vote game in his 100th AFL game, finishing equal-seventh in the 2010 Brownlow Medal count, and having the highest number of votes for a player from a team finishing outside the final eight. Watson was once again awarded the Crichton Medal, earning 291 points, which was 50 points clear of runner-up Heath Hocking. Watson had a relatively good season in 2011 despite missing six weeks with a hamstring injury, earning 15 Brownlow votes and finishing runner-up in the Crichton Medal, losing out to up-and-coming third-year midfielder David Zaharakis. Watson completed an outstanding 2012 season by winning the Brownlow Medal with 30 votes (though he was later ruled ineligible after being suspended during the Essendon Football Club supplements saga). Along with his third Crichton Medal, he also won a handful of other accolades, including the AFLPA best captain award, the Lou Richards Medal and selection in the 2012 All-Australian team as the centreman. Watson missed three weeks with a broken collarbone in 2013, but had another consistent season, earning 17 Brownlow votes, finishing runner-up in the Crichton Medal to former St Kilda utility (and later successor as captain) Brendon Goddard, and being named on the interchange in the 2013 All-Australian team. Between 2006 and 2013, Watson finished all but one season in the top two for votes for the Crichton Medal.
Drug cartels in Mexico are heavily involved in public relations and information warfare, employing tools such as food handouts, social media accounts, press release-style videos, narco corridos, and group chats through private messaging platforms primarily WhatsApp and Telegram. Cartel propaganda seeks to influence public opinion, threaten or discredit rivals, and coordinate between organizations. Physical "narco messages", ranging from printed banners to handwritten notes, are often displayed in public spaces or left at crime scenes. Some groups, notably the Jalisco New Generation Cartel (CJNG), maintain dedicated propaganda arms producing coordinated messages with logos, slogans, and professional formatting. Propaganda is also directed at cartel members themselves, with organizations such as La Familia Michoacana and the Knights Templar promoting religious, mythologizing narratives to reinforce loyalty. In 2011, President Felipe Calderón met with major media outlets, urging them to reduce sensationalist coverage and limit the dissemination of cartel messaging. Many cartels tacitly control local information environments by threatening journalists, bloggers, and others who speak out against them. In recent years, cartel messaging has moved into social media short-form videos and imagery that glamorize cartel life, weapons, and loyalty. Cartel slang is increasingly echoed in popular culture, blurring between criminal identity signifiers and everyday Mexican Spanish.

The main proposal is modulation of neurotrophic factors such as brain-derived neurotrophic factor, supported largely by animal experiments. Receptor-level targets have not been firmly established. Most reviews describe the mechanism as only partially characterized.
The compound is registered as a medicine in Russia, so clinical work has concentrated there. Trials elsewhere are few and generally small. This geographic concentration is a recognized limitation in evidence reviews.
Some fraction may reach the central nervous system through olfactory pathways, and this is often cited as the rationale for the nasal route. The size of that fraction in humans is not well quantified. Blood concentrations after nasal dosing are low, which complicates measurement.
It consists mainly of animal experiments and small clinical reports, with much of the clinical material published in Russian-language journals. Large independent trials are scarce. Separating reliable effects from chance findings is consequently difficult.
