This is a working overview of Semax, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-02-22 and is reviewed periodically as new material appears.
The compound was developed in the 1980s at the Institute of Molecular Genetics in Moscow, where it emerged from research on short ACTH fragments and their effects on the central nervous system. Russian pharmaceutical listings describe it as a nootropic and neuroprotective agent, most often formulated as nasal drops. It is not a marketed medicine in the United States or the European Union, and no pharmacopoeial monograph covers it. Consequently, most published clinical experience with the substance originates from a small number of research centres, mainly in Russia and neighbouring countries.
Pharmacological accounts link semax to melanocortin signalling and to modulation of neurotrophic factor expression, particularly brain-derived neurotrophic factor and nerve growth factor. Much of this evidence comes from rodent studies using intranasal delivery, a route chosen because it allows peptides to reach the central nervous system with limited systemic exposure. Whether the same mechanisms operate in humans at comparable magnitude remains an open question. The precise receptor or receptors responsible for the reported behavioural and neuroprotective effects have not been conclusively identified.
Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
The parent fragment ACTH(4-10) carries the sequence Met-Glu-His-Phe-Arg-Trp-Gly. Semax replaces the arginine and tryptophan positions with a proline-glycine-proline tail, giving Met-Glu-His-Phe-Pro-Gly-Pro. That change removes residues associated with adrenal stimulation, so the peptide does not drive cortisol release the way full ACTH does. This distinction shapes how the compound is grouped in the literature, where it sits with neuropeptides and peptide neuromodulators rather than with corticosteroids.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.

Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.
Clinical evidence consists largely of small trials with modest sample sizes, often without independent replication. Reported endpoints include cognitive scores, recovery after stroke, and visual function, but study designs vary widely and few trials meet contemporary reporting standards. Systematic reviewers have noted a high risk of bias in several of these reports. No large multicenter trial conducted outside Russia has been published. The compound is therefore best described as investigational in most jurisdictions, with its clinical role still unresolved.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its chain combines the first seven residues of corticotropin with a C-terminal proline-glycine-proline extension, a modification intended to slow enzymatic breakdown. The free peptide has a molecular mass near 813.9 daltons. It belongs to the class of ACTH-derived fragments studied for central nervous system activity rather than for adrenal steroid stimulation. This structural relationship to a natural hormone fragment is the usual starting point for describing the compound in the literature.
The proposed mechanism centres on neurotrophic signalling rather than direct receptor activation. Semax is reported to increase expression of brain-derived neurotrophic factor and nerve growth factor in several brain regions, and to shift the balance between excitatory and inhibitory neurotransmitter systems. Interaction with melanocortin receptors has been suggested because of the parent ACTH fragment. Many of these findings come from rodent studies, and the extent to which they translate to human physiology remains an open question.
Scientific literature on semax is unevenly distributed. A substantial share of published work originates from a small number of laboratories in Russia, while independent replication elsewhere is limited. Human data consist mostly of small trials with short follow-up, and several reported outcomes rely on subjective rating scales. Questions about how much intact peptide reaches the central nervous system after nasal administration, and how long it persists there, are still unresolved. The compound is best described as an active research subject rather than a settled pharmacological agent.

=== Proposed amalgamation === By early May 2026, several district and regional councils in Northland, Taranaki, the West Coast, Waikato, the Bay of Plenty, Wellington Wairarapa, Hawke's Bay and Southland Regions were discussing options to merge into unitary authorities as part of the Sixth National Government's policy of "simplifying" local government. On 5 May, the Local Government Minister Simon Watts and the RMA Reform Minister Chris Bishop issued local councils with a three-month timeframe to come up with amalgamation plans under the new "Head Start" approach. These amalgamation plans replaced the earlier proposed "combined territorial boards" and would be assessed by the newly-established Ministry for Cities, Environment, Regions and Transport (MCERT). The Government's 2026 amalgamation ultimatum received support from several local government leaders including Mayor of Nelson Nick Smith, Mayor of Southland Rob Scott, Mayor of Timaru Nigel Brown, Mayor of South Wairarapa Dame Fran Wilde, Mayor of Whangārei Ken Couper, Mayor of New Plymouth Max Brough, and Mayor of Invercargill Tom Campbell. By contrast, Mayor of Gore Ben Bell and Mayor of Rotorua Tania Tapsell expressed reservations about merger and the short time-frame. Meanwhile Mayor of Tasman Tim King preferred that the central government decide the local government model rather than delegating it to local councils.

Computing the total disintegration energy given by the equation Edi = (mi - mf - mp)c2 where mi is the initial mass of the nucleus, mf is the mass of the nucleus after particle emission, and mp is the mass of the emitted (alpha-)particle, one finds that in certain cases it is positive and so alpha particle emission is possible, whereas other decay modes would require energy to be added. For example, performing the calculation for uranium-232 shows that alpha particle emission releases 5.4 MeV of energy, while a single proton emission would require 6.1 MeV. Most of the disintegration energy becomes the kinetic energy of the alpha particle, although to fulfill conservation of momentum, part of the energy goes to the recoil of the nucleus itself (see atomic recoil). However, since the mass numbers of most alpha-emitting radioisotopes exceed 210, far greater than the mass number of the alpha particle (4), the fraction of the energy going to the recoil of the nucleus is generally quite small, less than 2%. Nevertheless, the recoil energy (on the scale of keV) is still much larger than the strength of chemical bonds (on the scale of eV), so the daughter nuclide will break away from the chemical environment the parent was in. The energies and ratios of the alpha particles can be used to identify the radioactive parent via alpha spectrometry. These disintegration energies, however, are substantially smaller than the repulsive potential barrier created by the interplay between the strong nuclear and the electromagnetic force, which prevents the alpha particle from escaping.
Twelve days later the 6th Division held a victory parade of its 1,200 guns, tanks and other vehicles at Monza racetrack, 16 kilometres (9.9 mi) north of Milan. The Rhodesians separated from their vehicles after the parade, then spent May and June 1945 as occupation troops in Lombardy before returning home.
=== Cataracts === A review measured serum tocopherol and reported higher serum concentration was associated with a 23% reduction in relative risk of age-related cataracts (ARC), with the effect due to differences in nuclear cataract rather than cortical or posterior subcapsular cataract. In contrast, meta-analyses reporting on clinical trials of alpha-tocopherol supplementation reported no statistically significant change to risk of ARC compared to placebo.

After a difficult few months at Eckartsau, the Imperial Family received aid from an unexpected source. Prince Sixtus had met King George V and appealed to him to help the Habsburgs. George was reportedly moved by the request, it being only months since his imperial relatives in Russia had been executed by revolutionaries, and promised "We will immediately do what is necessary." Several British Army officers were sent to help Charles, most notably Lieutenant-Colonel Edward Lisle Strutt, who was a grandson of Lord Belper and a former student at the University of Innsbruck. On 19 March 1919, orders were received from the War Office to "get the Emperor out of Austria without delay". With some difficulty, Strutt managed to arrange a train to Switzerland, enabling the Emperor to leave the country with dignity without having to abdicate. Charles, Zita, their children and their household left Eckartsau on 24 March escorted by a detachment of British soldiers from the Honourable Artillery Company under the command of Strutt.
One weakness of these measures is that they are not good indicators of liver vitamin A stores as retinyl esters in hepatic stellate cells. The amount of vitamin A leaving the liver, bound to retinol binding protein (RBP), is under tight control as long as there are sufficient liver reserves. Only when liver content of vitamin A drops below approximately 20 μg/gram will concentration in the blood decline.
Some that partake in looksmaxxing also look for others to rate their appearance, with some engaging in anonymous message boards associated with incel subculture. This rating, together with one's perceived status and wealth, has been considered to be one's "sexual market value" (also known as SMV). Those who look to determine someone's attractiveness usually check for a variety of facial features. One of the features checked for in men is "hunter eyes", which refers to a positive canthal tilt, little to no upper eyelid exposure and low-set eyebrows, resembling the eye area of a predatorial animal. Additional features checked for include hollow cheeks, a defined jawline and "pursed lips". In order to achieve these features, some perform acts such as the aforementioned "mewing", rubbing against the orbital area, or getting surgery. On some incel forums, users deemed below average may be harassed, including being told to take their own lives. Additional terms have been coined by these communities in an effort to "dominate others". These phrases include "mogging", which refers to asserting dominance over another person based on their appearance, and being "Y-pilled", which refers to one viewing themselves as more masculine than the other and is a spin on the phrase "redpilled". The term also comes from "AMOG", which is a new acronym for "alpha male of the group". In the 2020s, the smellmaxxing trend became an offshoot of looksmaxxing.

It is described as a synthetic analogue of the ACTH(4–10) fragment, a short segment of adrenocorticotropic hormone. Its sequence differs from that fragment and includes two proline residues, which influence stability and behaviour in solution.
It appears in Russian pharmaceutical listings as a nasal formulation, but it is not an authorised medicine in the United States or the European Union. Outside those markets it is normally encountered as a research chemical rather than a prescription product.
Laboratory and animal work points to melanocortin signalling and changes in neurotrophic factor levels, especially brain-derived neurotrophic factor. The exact receptor targets and the degree to which these findings transfer to humans are still unresolved.
Registered medical products are most often 0.1% nasal drops. Research suppliers ship lyophilized powder, usually in milligram quantities, which is dissolved before use. The active peptide is the same in both cases; presentation and excipients differ.
