This is a working overview of Pro-Gly-Pro tail, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2025-08-17 and is reviewed periodically as new material appears.
The parent fragment ACTH(4-10) carries the sequence Met-Glu-His-Phe-Arg-Trp-Gly. Semax replaces the arginine and tryptophan positions with a proline-glycine-proline tail, giving Met-Glu-His-Phe-Pro-Gly-Pro. That change removes residues associated with adrenal stimulation, so the peptide does not drive cortisol release the way full ACTH does. This distinction shapes how the compound is grouped in the literature, where it sits with neuropeptides and peptide neuromodulators rather than with corticosteroids.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
Scientific literature on semax is unevenly distributed. A substantial share of published work originates from a small number of laboratories in Russia, while independent replication elsewhere is limited. Human data consist mostly of small trials with short follow-up, and several reported outcomes rely on subjective rating scales. Questions about how much intact peptide reaches the central nervous system after nasal administration, and how long it persists there, are still unresolved. The compound is best described as an active research subject rather than a settled pharmacological agent.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It was derived from the ACTH(4-10) fragment, a short segment of adrenocorticotropic hormone that lacks the hormonal activity associated with the full-length peptide. Researchers at the Institute of Molecular Genetics in Moscow developed the compound during the 1980s. It has been registered as a pharmaceutical product in Russia and several neighbouring countries, where it is supplied as a nasal solution, and it is also sold internationally as a research chemical.
The proposed mechanism centres on neurotrophic signalling rather than direct receptor activation. Semax is reported to increase expression of brain-derived neurotrophic factor and nerve growth factor in several brain regions, and to shift the balance between excitatory and inhibitory neurotransmitter systems. Interaction with melanocortin receptors has been suggested because of the parent ACTH fragment. Many of these findings come from rodent studies, and the extent to which they translate to human physiology remains an open question.

Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.
Regulatory status differs sharply between jurisdictions. In Russia the peptide is registered as a prescription nasal preparation, while agencies such as the United States Food and Drug Administration have not approved it for any indication. Products sold elsewhere are typically labeled for laboratory research only, and such labels shift responsibility for safe handling to the purchaser. Because the same name covers pharmaceutical-grade nasal drops and bulk research powder, identity and purity documentation becomes the main practical concern when comparing sources.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.

Most neptunium (and plutonium) now encountered in the environment is due to atmospheric nuclear explosions that took place between the detonation of the first atomic bomb in 1945 and the ratification of the Partial Nuclear Test Ban Treaty in 1963. The total amount of neptunium released by these explosions and the few atmospheric tests that have been carried out since 1963 is estimated to be around 2500 kg. The overwhelming majority of this is composed of the long-lived isotopes 236Np and 237Np since even the moderately long-lived 235Np (half-life 396 days) would have decayed to less than one-billionth (10−9) its original concentration over the intervening decades. An additional very small amount of neptunium, produced by neutron irradiation of natural uranium in nuclear reactor cooling water, is released when the water is discharged into rivers or lakes. The concentration of 237Np in seawater is approximately 6.5 × 10−5 millibecquerels per liter: this concentration is between 0.1% and 1% that of plutonium. Once released in the surface environment, in contact with atmospheric oxygen, neptunium generally oxidizes fairly quickly, usually to the +4 or +5 state. Regardless of its oxidation state, the element exhibits much greater mobility than the other actinides, largely due to its ability to readily form aqueous solutions with various other elements. In one study comparing the diffusion rates of neptunium(V), plutonium(IV), and americium(III) in sandstone and limestone, neptunium penetrated more than ten times as well as the other elements.

== Catalytic mechanism == The pyridoxal-5′-phosphate-dependent enzymes (PLP enzymes) catalyze myriad reactions. Although the scope of PLP-catalyzed reactions appears to be immense, the unifying principle is the formation of an internal lysine-derived aldimine. Once the amino substrate interacts with the active site, a new Schiff base is generated, commonly referred to as the external aldimine. After this step, the pathway for each PLP-catalyzed reactions diverge.
=== Conceptual development === Ilyushin began work on the aerobus in late 1969, initially by assessing the development potential of existing aircraft. An enlarged Il-62 (the Il-62-250) would have had a 30-tonne payload, 259 seats and a 6.8 metre/22 ft longer fuselage: a virtual analogue of the Douglas DC-8 "Super Sixty" series. Other proposed Il-62 modifications involved double-deck and "two fuselages side-by-side" developments. There was also a project to "civilianise" the Il-76. From March 1970 the bureau developed all-new designs under the Il-86 designation. Instead of the "appropriate technology" approach of the Il-62, these designs were to have powered controls, complex high-lift devices and advanced automation which would reduce the number of flightdeck crew. An early avanproyekt was shown to the Soviet leadership at an exhibition of civil aviation innovations at Vnukovo-2 Airport near Moscow on May 17, 1971. A scale model with the designation of "Il-86" showed the "self-loading" concept with integral boarding stairs, below-deck luggage stores, and below-deck midships galley. It had a twin-aisle interior with nine-abreast seating in a "3–3–3" layout. Ilyushin considered it politic to make the interior wider than any planned airliner except the Boeing 747. The 6.07 m (19.9 ft) fuselage diameter was partly dictated by the need to provide standing room in the underfloor luggage compartments. The Il-86 had the second-widest fuselage of any airliner until the Boeing 777. On this basis, on 9 March 1972, the bureau was asked to proceed with detailed design.

== Contributions == De made significant contributions to the recent understanding of cholera and related diarrhoeal diseases and set a permanent milestone in the modern view of diseases caused by bacterial exotoxins. Followed by the discovery of Vibrio cholerae in 1884 by Robert Koch, many works have been carried out all over the world to answer many questions related with its pathogenesis and mode of transmission in causing outbreaks. The seminal works of De in Calcutta (now Kolkata) during 1950–60 breached several qualms pertaining to the enteric toxin produced by bacteria including V. cholerae and Escherichia coli. Three of his works viz., ligated intestinal loop method (which was a reinvention of Violle and Crendiropoulo method in 1915, but De was unaware of this work and made an independent discovery) for studying cholera in rabbit model; ileal loop model to demonstrate the association of some strains of E. coli with diarrhoea and lastly but most importantly is his discovery of cholera toxin in 1959 in the cell-free culture filtrate of V. cholerae that stimulated a specific cellular response. As noted by Garfield in his 1986 tribute, De was the first to demonstrate that cholera bacteria secrete enterotoxin. This discovery eventually promoted research to find a treatment aimed directly at neutralising the cholera enterotoxin. De's paper "Enterotoxicity of bacteria-free culture-filtrate of Vibrio cholerae," while initially unrecognised, today is considered a milestone in the history of cholera research. Biochemist W. E.
=== Prevalence in rain, soil, water bodies, and air === In 2022, levels of at least four perfluoroalkyl acids (PFAAs) in rain water worldwide greatly exceeded the EPA's lifetime drinking water health advisories as well as comparable Danish, Dutch, and European Union safety standards, leading to the conclusion that "the global spread of these four PFAAs in the atmosphere has led to the planetary boundary for chemical pollution being exceeded". The most common PFAS found in the environment is Trifluoroacetic acid (TFA). Its presence is ubiquitous in the environment, especially in aquatic ecosystems, where it persists with increasing concentrations globally. It had been thought that PFAAs would eventually end up in the oceans, where they would be diluted over decades, but a field study published in 2021 by researchers at Stockholm University found that they are often transferred from water to air when waves reach land, are a significant source of air pollution, and eventually get into rain. The researchers concluded that pollution may impact large areas. Soil is also contaminated and the chemicals have been found in remote areas such as Antarctica. Soil contamination can result in higher levels of PFAS found in foods such as white rice, coffee, and animals reared on contaminated ground. In 2024, a worldwide study of 45,000 groundwater samples found that 31% of samples contained levels of PFAS that were harmful to human health; these samples were taken from areas not near any obvious source of contamination.
The Russian interior ministry placed Zelenskyy in its list of wanted criminals. Ukraine claimed to have shot down a Russian Su-25 fighter jet over Donetsk Oblast. The Russian defence ministry claimed to have shot four ATACMS missiles over Crimea. A court in Russia sentenced a Ukrainian POW captured during the Siege of Mariupol to 18 years imprisonment for membership in the Azov Battalion.
The most common tests performed by medical laboratory scientists are complete blood count (CBC), comprehensive metabolic panel (CMP), electrolyte panel, liver function tests (LFT), renal function tests (RFT), thyroid function test (TFT), urinalysis, coagulation profile, lipid profile, blood type, genotype, pack cells volume (PCV), hepatitis (HbsAg and HCV), glucose test (fasting blood sugar {FBS} and random blood sugar {RBS}), Endo cervical swab (ECS), high vaginal swab (HVS), semen fluid analysis (for fertility and post-vasectomy studies), serological studies and routine cultures. In some facilities that have few phlebotomists, or none at all, (such as in rural areas) medical laboratory scientists may perform phlebotomy. Because medical laboratory scientists have many transferable technical skills, employment outside of the medical laboratory is common. Many medical laboratory scientists are employed in government positions such as the FDA, USDA, non-medical industrial laboratories, and manufacturing. In the United Kingdom and the United States, senior laboratory scientists, who are typically post-doctoral scientists, take on significantly greater clinical responsibilities in the laboratory. In the United States these scientists may function in the role of clinical laboratory directors, while in the United Kingdom they are known as consultant clinical scientists.

Registered medical products are most often 0.1% nasal drops. Research suppliers ship lyophilized powder, usually in milligram quantities, which is dissolved before use. The active peptide is the same in both cases; presentation and excipients differ.
No. The peptide is synthetic, although it is modelled on a fragment of a naturally occurring hormone. ACTH(4-10) is generated when ACTH is cleaved, but the Pro-Gly-Pro extension is not a known endogenous sequence. It is therefore a designed analogue rather than a natural metabolite.
The label reflects reported effects on attention, memory and recovery after neurological injury in regional clinical reports. Those reports are mostly small and have not been widely replicated outside the region. The classification is descriptive and does not imply a single defined mechanism.
Semax is based on the ACTH(4-10) fragment, a seven-amino-acid segment of adrenocorticotropic hormone. The synthetic peptide retains the core sequence while removing regions associated with endocrine activity. This modification is intended to isolate effects on the nervous system.
