This is a working overview of freeze-thaw, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-01-07 and is reviewed periodically as new material appears.
Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.
Regulatory status differs sharply between jurisdictions. In Russia the peptide is registered as a prescription nasal preparation, while agencies such as the United States Food and Drug Administration have not approved it for any indication. Products sold elsewhere are typically labeled for laboratory research only, and such labels shift responsibility for safe handling to the purchaser. Because the same name covers pharmaceutical-grade nasal drops and bulk research powder, identity and purity documentation becomes the main practical concern when comparing sources.
Semax is a synthetic seven-amino-acid peptide whose sequence extends the ACTH(4-10) fragment with a C-terminal proline-glycine-proline tripeptide. The commonly cited sequence is Met-Glu-His-Phe-Pro-Gly-Pro, giving a molecular formula near C37H51N9O10S and a molecular weight close to 813.9 g/mol. It belongs to the broader class of synthetic ACTH fragments studied for central nervous system effects rather than for adrenal steroid stimulation. In practice the material appears as a lyophilized white powder for laboratory work or as a dilute saline solution in clinical settings.
Identity and purity are commonly assessed by reversed-phase high-performance liquid chromatography, with ultraviolet detection near 214 nanometers for the peptide backbone. Mass spectrometry, either electrospray or matrix-assisted laser desorption, confirms molecular mass and detects truncation or modification products. Amino acid analysis can verify composition. Because the sequence contains no strongly absorbing aromatic residue apart from phenylalanine, detection wavelengths are chosen carefully. Purity values above 95 percent are typical for research-grade material.
Quality documentation for research-grade peptide usually includes a certificate of analysis stating purity, identity, and residual counterion content. Batch-to-batch variation in purity and salt form is a recognized issue, and comparisons across suppliers require attention to the exact counterion, for example acetate versus trifluoroacetate. Sequence verification by tandem mass spectrometry provides stronger evidence than a single mass measurement. For any study reporting biological results, the analytical method and the measured purity should be stated explicitly. Independent verification of supplier claims is considered good practice.
Semax is supplied as a lyophilized powder that appears white to off-white. It dissolves readily in water, phosphate-buffered saline, and other aqueous media, which simplifies preparation of working solutions for laboratory use. The nasal products registered in Russia are dilute aqueous solutions, typically around 0.1 percent peptide by weight. Organic solvents are rarely necessary and can complicate handling. Because the peptide is hygroscopic, weighing should be performed quickly and with minimal exposure to ambient humidity.

The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
The parent fragment ACTH(4-10) carries the sequence Met-Glu-His-Phe-Arg-Trp-Gly. Semax replaces the arginine and tryptophan positions with a proline-glycine-proline tail, giving Met-Glu-His-Phe-Pro-Gly-Pro. That change removes residues associated with adrenal stimulation, so the peptide does not drive cortisol release the way full ACTH does. This distinction shapes how the compound is grouped in the literature, where it sits with neuropeptides and peptide neuromodulators rather than with corticosteroids.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Freeze-dried foods became a major component of astronaut and military rations. What began for astronaut crews as tubed meals and freeze-dried snacks that were difficult to rehydrate, were transformed into hot meals by improving the process of rehydrating freeze-dried meals with water. As technology and food processing improved, NASA looked for ways to provide a complete nutrient profile while reducing crumbs, disease-producing bacteria, and toxins. The crumb problem was solved with the addition of a gelatin coating on the foods to lock in and prevent crumbs. The complete nutrient profile was improved with the addition of an algae-based vegetable-like oil to add polyunsaturated fatty acids. Polyunsaturated fatty acids are beneficial in mental and vision development and, as they remain stable during space travel, can provide astronauts with added benefits. Disease-producing bacteria and toxins were reduced by quality control and the development of the Hazard Analysis and Critical Control Points (HACCP) plan, which is widely used today to evaluate food material before, during, and after processing. With the combination of these three innovations, NASA could provide safe and wholesome foods to their crews from freeze-dried meals. Military rations have also come a long way, from being served cured pork and corn meal to beefsteaks with mushroom gravy. How rations are chosen and developed is based on acceptance, nutrition, wholesomeness, producibility, cost, and sanitation.
=== Media === The Houthis are said to have "a huge and well-oiled propaganda machine". They have established "a formidable media arm" with the Lebanese Hezbollah's technical support. The format and content of the group's leader, Abdul-Malik al-Houthi's televised speeches are said to have been modeled after those of Hezbollah's Secretary General, Hassan Nasrallah. Following the peaceful youth uprising in 2011, the group launched its official TV channel, Almasirah. The group operates up to 25 print and electronic newspapers, along with various online news services. One of the most versatile form of Houthi mass media are the zawamil, a genre of primarily tribal oral poetry embedded in Yemen's social fabric. The zamil, rooted in cultural tradition, has been weaponised by the Houthis as a tool of propaganda and remains one of the most popular and rapidly growing platforms of Houthi propaganda, sung by popular vocalists like Issa al-Laith and disseminated through various social media platforms including YouTube, Twitter and Telegram. The Spectator describes Houthi zawamil as its most successful part of their propaganda, stressing the movement's claimed virtues of piety, bravery and poverty in comparison with the corruption, wealth and hypocrisy of their adversaries, the Saudi-led coalition, and Arab states allied to Israel. The Houthis use radio as an effective tool for spreading influence, often seizing stations and confiscating equipment from outlets that fail to comply with their broadcast restrictions.
== False claims == Since around 2008, GcMAF has been promoted as a cure for cancer, HIV, autism and other conditions. Three out of four of the original studies authored by Yamamoto (published between 2007 and 2009) were retracted by the scientific journals in which they were published in 2014, officially due to irregularities in the way ethical approval was granted. Retraction reasons also included methodological errors in the studies. The integrity of the research, conducted by Nobuto Yamamoto and colleagues, that originally prompted claims regarding cancer and HIV has been questioned. The UK Medicines and Healthcare products Regulatory Agency and Cancer Research UK has warned the public about spurious claims of clinical benefits, misleadingly based on reduced levels of the alpha-N-acetylgalactosaminidase enzyme (also known as nagalase), whose production might be increased in many cancers. In 2014 the Belgian Anticancer Fund communicated serious concerns about published studies on GcMAF by Yamamoto and colleagues. In 2015 the UK Medicines and Healthcare products Regulatory Agency (MHRA) closed a factory in Milton, Cambridgeshire owned by David Noakes' company Immuno Biotech that manufactured GcMAF for cancer treatment. In September 2018 Noakes pleaded guilty in UK to manufacturing a medicinal product without a manufacturer's licence, selling or supplying medicinal products without market authorisation, and money laundering, and sentenced to 15 months of jail.
== Mechanism of action == GLP-1 receptors improve insulin sensitivity and increase satiety. Retatrutide has shown to increase insulin secretion. Furthermore, agonism of the glucagon receptor increases energy expenditure and promotes fat loss and metabolic activity. With current research, activation of these receptors has overall reduced caloric intake and increased energy expenditure, successfully leading to higher weight loss compared to alternative treatments.
=== Alternative medicine === Several alternate therapies such as vitamin E treatment have been studied, though without control groups. Most doctors do not value those treatments. None of these treatments stops or cures the condition permanently. A 1949 study of vitamin E therapy found that "In twelve of the thirteen patients there was no evidence whatever of any alteration. ... The treatment has been abandoned." "Cold" laser treatment or "photomodulation" (using red and infrared at low power) was informally discussed in 2013 at an International Dupuytren Society forum, as of which time little or no formal evaluation of the techniques had been completed. The term "photobiomodulation" has expanded to include other low-power light sources such as blue LEDs, making it a form of light therapy rather than specifically a laser-based treatment. No human trials have been performed yet. In 2021 improvement of Dupuytren's disease in a single patient by ablative laser surgery with a fractionated 10.6 μm carbon-dioxide laser was reported. This is different from a cold laser: in laser surgery, tissue is physically removed by heat by photoablation (also known as "laser blasting").

=== Brand names === Propranolol was first marketed under the brand name Inderal, manufactured by ICI Pharmaceuticals (now AstraZeneca), in 1965. "Inderal" is a quasi-anagram of "Alderlin", the trade name of pronethalol (which propranolol replaced); both names are an homage to Alderley Park, the ICI headquarters where the drugs were first developed. Propranolol is also sold under the brand names Avlocardyl, Deralin, Dociton, Hemangeol, Inderalici, InnoPran XL, Indoblok, Sumial, Anaprilin, and Bedranol SR (Sandoz). In India, it is sold under brand names such as Ciplar and Ciplar LA by Cipla.
8969, which declared the third Friday of August a special holiday in Davao City. He expressed disinterest with his new post, describing it as less engaging compared to his time as mayor. He attempted to resign in 1999 after his son Paolo was accused of mauling a hotel guard, but House Speaker Manny Villar and President Joseph Estrada declined his resignation. After embarking on a diplomatic trip to Brazil between 2000 and 2001, Duterte made a brief stopover in the United States, but as he was about to step out of the Los Angeles International Airport, he was held up by law enforcement officers and interrogated about his travel documents. After the stopover, Duterte never again returned to the United States. In 2001, he was elected mayor of Davao for a fourth unconsecutive term, defeating his former ally, incumbent Mayor Benjamin de Guzman; his estranged sister Jocelyn attempted to run against him as well, but was disqualified by the Commission on Elections. He was re-elected in 2004, defeating de Guzman for the second time, and in 2007 with virtually no opposition. In 2003, Duterte formally laid out the scope of Davao City's Chinatown, the first Chinatown in Mindanao, along the newly renamed Magsaysay Street, while establishing the Davao City Chinatown Development Council to oversee the area's development.
Biological devolution or de-evolution is a misnomer, not only because it implies that organisms can only evolve backward or forward, but also because it implies that evolution may cause organisms to evolve in the "wrong" direction. The phrase "survival of the fittest" refers to biological fitness, not physical fitness. Biological fitness is the quantitative measure of individual reproductive success, e.g. the tendency of lineages containing individuals that produce more offspring in a particular environment to persist and thrive in that environment. Further, while the related concepts of "survival of the fittest" and "natural selection" are often used interchangeably, they are not the same: natural selection is not the only form of selection that determines biological fitness (see sexual selection, fecundity selection, viability selection, and artificial selection). Evolution does not "plan" to improve an organism's fitness to survive. This misconception is encouraged as it is common shorthand for biologists to speak of a purpose as a concise form of expression (sometimes called the "metaphor of purpose"); it is less cumbersome to say "Dinosaurs may have evolved feathers for courtship" than "Feathers may have been selected for when they arose as they gave dinosaurs a selective advantage during courtship over their non-feathered rivals". However, this can result in many students explaining evolution as an intentional and purposeful process. Mutations are not all equally likely, nor do they occur at the same frequency everywhere in the genome.
=== Bone marrow barrier === The blood vessels of the bone marrow constitute a barrier, inhibiting immature blood cells from leaving the marrow. Only mature blood cells contain the membrane proteins, such as aquaporin and glycophorin, that are required to attach to and pass the blood vessel endothelium. Hematopoietic stem cells may also cross the bone marrow barrier, and may thus be harvested from blood.

It is a short synthetic peptide built from seven amino acids: methionine, glutamic acid, histidine, phenylalanine and three prolines. The sequence derives from the 4-10 fragment of adrenocorticotropic hormone with an added proline-glycine-proline tail. No plant or animal extract is involved; the material is produced by solid-phase peptide synthesis.
It was developed in the Soviet Union during the early 1980s by groups at the Institute of Molecular Genetics in Moscow. Russian approval for intranasal use followed, and it has been marketed there since. Western laboratories encountered it mainly through translated literature and, later, through online research-chemical trade.
It holds a Russian registration as a prescription nasal product but has no approval from the European Medicines Agency or the United States Food and Drug Administration. Buyers outside Russia usually receive material sold strictly for laboratory research. That distinction matters because research-grade and pharmaceutical-grade products carry different documentation and testing expectations.
Solid peptide is best kept frozen at about minus 20 degrees Celsius in a sealed container with desiccant. It should be allowed to reach room temperature before opening to prevent condensation. Repeated warming and cooling of the same vial is discouraged.
