=== Atomic and molecular physics === Some atomic energy levels are metastable. Rydberg atoms are an example of metastable excited atomic states. Transitions from metastable excited levels are typically those forbidden by electric dipole selection rules. This means that any transitions from this level are relatively unlikely to occur. In a sense, an electron that happens to find itself in a metastable configuration is trapped there. Acetylcysteine was patented in 1960 as a mucolytic agent. In the 1960s, it was used for managing the mucous symptoms of illnesses including cystic fibrosis, asthma, and chronic bronchitis, as well as other uses, such as a chelating agent in gold therapy. In the late 1960s, the popularity of acetaminophen led to an increase in overdose hepatotoxicity. In 1974, it was discovered that the mechanism was oxidation via cytochrome P450, and administering glutathione-like reducing agents, such as IV cysteine, or oral methionine, were successful in preventing hepatotoxicity. In the US, the then-common formulation of NAC was not certified as pyrogen-free, and thus not approved for IV use. The producer, Mead Johnson, was unwilling to take the expense of certification. In 1977, there was the first case report of oral NAC preventing acetaminophen overdose hepatoxicity in the US. Meanwhile, in the UK, IV NAC (trade name, Parvolex) was found to be more effective and has less side effect than IV cysteamine (associated with a "general feeling of misery") or methionine. There was a dispute concerning whether the oral NAC and the IV NAC is more effective, with the conclusion reached in 1999, that they are roughly equally effective. Another producer finally obtained approval for the IV form of NAC (trade name, Acetadote) in the US in 2004. Amazon removed acetylcysteine for sale in the US in 2021, due to claims by the Food and Drug Administration (FDA) of it being classified as a drug rather than a supplement.

=== Heptapeptide backbone synthesis === The heptapeptide backbone of teicoplanin is assembled by the nonribosomal peptide synthetases (NRPSs) TeiA, TeiB, TeiC and TeiD. Together these comprise seven modules, each containing a number of domains, with each module responsible for the incorporation of a single amino acid. Modules 1, 4, and 5 activate L-4-Hpg as the aminoacyl-AMP, modules 2 and 6 activate L-Tyr, and modules 3 and 7 activate L-3,5-Dpg. The activated amino acids are covalently bound to the NRPS as thioesters by a phosphopantetheine cofactor, which is attached to the peptidyl carrier protein (PCP) domain. The enzyme bound amino acids are then joined by amide bonds by the action of the condensation (C) domain. The heptapetide of teicoplanin contains 4 D-amino acids, formed by epimerization of the activated L-amino acids. Modules 2, 4 and 5 each contain an epimerization (E) domain which catalyzes this change. Module 1 does not contain an E domain, and epimerization is proposed to be catalysed by the C domain. In all, six of the seven total amino acids of the teicoplanin backbone are composed of nonproteinogenic or modified amino acids. == Injectable materials == Many different materials have been used as perianal injectable bulking agents. The ideal injectable or implantable material would be biocompatible, non-migratory, non-allergenic, non-carcinogenic and non-immunogenic (and therefore induce a minimal inflammatory and fibrotic reaction). The material should also be easy to inject. The particles should be greater than 80 μm in diameter in order to prevent migration away from the injection site. On the other hand, materials with particles small enough to be used in small caliber needles may be desirable, in order to leave a smaller needle track, which may reduce the chance of leakage of the material via the needle track. Alternatively, some materials are shape-retaining porous hydrogels with no particles. The ideal material should produce an improvement in continence not only in the short term, but in the long term, and repeated procedures should not be necessary.
